The Phosphate Buffer for Spectrophotometry is used to adjust laboratory water to a pH of 8.4 – 8.8. Adjusting water used to dilute RNA or DNA samples for spectrophotometry to a slightly alkaline pH greatly improves UV measurements and increases
sensitivity for the spectrophotometric detection of proteins contaminating nucleic acid preparations. The Phosphate Buffer contains 0.3 M Na2HPO4 that is diluted to make a working solution of 1 – 3 mM Na2HPO4.
